Casson studied at Eastbourne College and Cambridge (1929-1931), followed by the Bartlett School of Architecture. He taught architecture at Cambridge until 1939, when he worked in the Camouflage Unit. In 1948, Casson was appointed Director of Architecture for the Festival of Britain, after which he was knighted. In partnership with Neville Conder, Casson designed the Elephant House at London Zoo, Staff House, University of Birmingham, and the Arts Faculty for Cambridge. He was Professor of Interior Design at the Royal College of Art (1955-77) and Provost from 1970. As President of the Royal Academy (1976- 84) he saw its development as the most successful exhibition venue in London.
